The research program pursued by Antonio Iavarone aims to decipher the biological and genetic changes that drive subgroups of malignant brain tumors and to use this information for rational therapeutic stratification. The Iavarone group identified major regulators of cancer development and progression in various subgroups of brain tumors. They discovered the first example of oncogenic and tumor-addicted gene fusions in glioblastoma (FGFR3-TACC3) and reported that FGFR3-TACC3 fusions trigger tumor development by activating oxidative phosphorylation. These fusions are among the most common chromosomal translocations in all types of human cancer, and the FDA cleared the targeting of these chromosomal translocations with FGFR inhibitors. Antonio Iavarone was chairman of the Pan-Glioma ATLAS-TCGA Working Group and Neurofibromatosis 1 Synodos, which brought together international researchers to formulate guidelines for the accurate diagnosis and prognosis of glioma patients. Using all available computational and experimental tools, they combine innovation in both areas to continue to make transformative mechanistic discoveries and provide personalized therapeutics to an increasing number of patients with lethal tumor types.
